Help with slow computers

I started working for a very small non-profit recently and have just enough knowledge of computers to be dangerous. But my knowledge FAR eclipses anyone else around here. I have "fixed" two of the computers in the company and they are running like new machines. My boss' computer is EXTREMELY slow even though it seems to have adequate disk space. We have no budget at present to hire someone from outside. Can anyone give me some pointers on where I can start?
